Изучить распространенность полиморфизмов rs17549193 и rs7851696 гена фиколина-2 FCN2 среди ненцев, долган-нганасан и славян с помощью молекулярно-генетического исследования однонуклеотидных полиморфизмов.</p></sec><sec><title>Дизайн</title><p>Дизайн. Сравнительное популяционно-генетическое исследование.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Обследованы новорожденные Таймырского Долгано-Ненецкого района Красноярского края — ненцы (n = 261) и долганы-нганасаны (n = 104) — и дети-славяне г. Красноярска 8–18 лет с бронхиальной астмой разной степени тяжести и без нее. Проведена экстракция ДНК из венозной крови и сухих пятен крови. Генотипирование полиморфизмов rs17549193 и rs7851696 гена FCN2 было произведено методом полимеразной цепной реакции в реальном времени.</p></sec><sec><title>Результаты</title><p>Результаты. Минорный аллель полиморфизма rs17549193 гена FCN2 у ненцев встречается значимо реже, чем у славян группы контроля и больных астмой, — 19,9% против 28,8% (р &lt; 0,001) и 27,5% (р = 0,002). Распространенность аллельного варианта Т полиморфного локуса rs7851696 FCN2 у ненцев и долган-нганасан также меньше, чем у славян с астмой и без нее, — 7,3 и 3,8% против 14,6 и 12,6% (р &lt; 0,05).</p></sec><sec><title>Заключение</title><p>Заключение. В связи с тем, что большинство заболеваний человека обладают генетической детерминированностью, актуальным является изучение характерных особенностей в отдельных популяциях для составления рекомендаций организациям здравоохранения. Понимание роли и функций фиколина-2 как одного из протеинов лектинового пути активации системы комплемента имеет большое значение для разработки новых методов профилактики заболеваний и оценки функционирования иммунной защиты.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Aim</title><p>Aim. To study the prevalence of polymorphisms rs17549193 and rs7851696 of the ficolin-2 gene FCN2 among the Nenets, Dolgan-Nganasan and Slavic populations using a molecular genetic study of single nucleotide polymorphisms.</p></sec><sec><title>Design</title><p>Design. Comparative population-genetic research.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. Newborn infants of the Taimyr Dolgan-Nenets district of the Krasnoyarsk Territory — Nenets (n = 261) and Dolgan-Nganasan (n = 104) — and Slavic children of Krasnoyarsk aged 8–18 years with and without bronchial asthma of varying severity were examined. DNA extraction from venous blood and dry blood stains was performed. The genotyping of polymorphisms rs17549193 and rs7851696 of the FCN2 gene was performed by real-time polymerase chain reaction.</p></sec><sec><title>Results</title><p>Results. The minor allele of the polymorphism rs17549193 FCN2 in Nenets is significantly less than in Slavs of control group and with asthma — 19.9% vs. 28.8% (p &lt; 0.001) and 27.5% (p = 0.002). The prevalence of the T allelic variant rs7851696 FCN2 in the Nenets and Dolgan-Nganasans is also lower than in the Slavs with and without asthma — 7.3 and 3.8% vs. 14.6 and 12.6% (p &lt; 0.05).</p></sec><sec><title>Conclusion</title><p>Conclusion. Due to the fact that most human diseases are genetic determinism, it is relevant to study the characteristic features in individual populations to draw up recommendations to healthcare organizations.
